Icariin promotes osteogenic differentiation of rat bone marrow stromal cells in vitro
Epimedium have effective function to cure the osteoporosis and promote fracture healing. As we know the
icariin ( include icariin 10%, icariin 20%, icariin 40%, icariin 50% icariin 60%, icariin 90% and icariin 98% )
is the most rich nutriton of flavonoid glycosides in epimedium. Chen Ke Ming group study show that the
icariin can inhibit osteoclastic absorption activity in vitro and improve the quantity of mineralized nodule and
activity of alkaline phosphatase on mesenchymal stem cells. The same study report from Yin Xue Mei group
found that the icariin should be the mainly effective activity to improve the function of bone strong in
Now ZhaiYunKung group from Lanzhou military region general hospital mainly to study the influence of related
gene expression factor in 4 kinds of mature bone from icariin through the method of fluorescent quantitation
(PCR). Method: rBMSCs were cultured by adherence screening method. Icariin was supplemented into the
culture at 1×10-5 mol·L-1. The osteogenic differentiation markers including alkaline phosphatase (ALP) activity,
CFU-FALP and mineralized bone modulus were compared between the icariin-supplemented group and the
control group. Total RNA was isolated and the gene expression of bFGF, IGF-1, Osterix(OSX) and Runx-2 was
investigated by RT Real-time PCR.
From above study, the researchers mainly found the icariin can improve the ALP activity, Osterix （OSX) amounts
and mineralized modulus significantly. Also it can enhance the mRNA level of bFGF, IGF-1, Osterix and Runx-2.
Then we can get the conclusion from the the study that Icariin enhances the osteogenic differentiation of rBMSCs
significantly, which suggested that icariin has the potentiality to be a new drug of anti-osteoporosis or fracture healing.
